The purpose of the study was to investigate the efficacy of Flavonoid fraction of Ixora coccinea (L.) in reducing the oxidative damage and inflammation in adjuvant induced rat models. The preliminary screening of percentage paw edema inhibition by aqueous and methanolic extracts of Ixora coccinea in carrageenan induced rats were done. The percentage inhibition result showed that aqueous and methanolic extract of Ixora had maximum effect. A dose dependant study was done with different doses of ICFF such as 1mg/kg, 2mg/kg, 5mg/kg and 10mg/kg. 5mg/kg body weight ICFF retained the normal oxidation status. So 5mg/kg dose was selected for further studies in chronic inflammatory model using Freund's Complete Adjuvant (FCA). For the study the rats were divided into four groups- A control group, Adjuvant alone (1mg/kg body weight), Adjuvant +ICFF (5mg/kg body weight), Adjuvant+ Indomethacin (3mg/kg body weight). The results of the study suggest that flavonoid fraction of Ixora coccinea inhibit the inflammatory mediators like COX, LOX, NOS and MPO thereby decreasing the inflammation induced by an adjuvant. The administration of ICFF also down regulated the transcription factor NF-kB and thereby the expression of cytokines TNF-α and IL-6 in rats. Thus, the present study suggests that ICFF has potential anti-inflammatory activity. Hence the flavonoid fraction can be used as promising anti-inflammatory drug. |
